microRNA let-7i-5p aggravates kidney fibrosis via targeting GALNT1

Renal fibrosis poses critical health problem. We aimed to investigate role of let-7i-5p in renal fibrosis. In silico reproduction Mouse Kidney FibrOmics browser was used identify potential target let-7i-5p. vivo validation conducted C57BL/6 mice with unilateral ureteral obstruction (UUO) and folic acid (FA) induction. vitro performed transforming growth factor (TGF)-β1-treated HK-2 cells. Mimics inhibitors let-7i-5p, gene polypeptide N-acetylgalactosaminyltransferase 1 (GALNT1) were monitored by RT-PCR Western blotting. Fibrosis markers, injury house-keeping genes evaluated. Levels interleukin (IL)-6, IL-1β, tumor necrosis (TNF)-α serum media measured ELISA. analysis showed gradual increase decrease GALNT1 over time the combination validated both mouse human miR-gene prediction databases. Expression decreased while expression increased tissues UUO FA mice. Serum IL-6, TNF-α levels elevated vivo. models revealed negative correlation between GALNT1.
